Problem!

If two rat snakes are housed together all year round, is it possible for just one of them to get scale rot?

Yes. If you have 2 kids sharing the same room is it possible for just one of them to get sick? I would reccomend housing them seperately except for breeding purposes & if scale rot is present the humidity is too high or they are being kept in too wet/dirty of a substrate (more likely cause). Husbandry must be changed to correct the problems that caused the scale rot & the specimens need to be seperated & the one treated accorddingly. Mark
